Freezer Medium for cDNA clones
Optimized for growth in microtiter plates

Make in 3 parts to avoid problems with
precipitation during autoclaving:.
Solution 1:
Tryptone 16g
Yeast Extract 10g
NaCl 5g
Dissolve in H2O, adjust pH
to 7.4 with NaOH, bring to final volume of 900 ml with H2O,
autoclave.
Solution 2: (This is a concentrated stock)
KH2PO4
17.96g
K2HPO4
62.7g
bring to 150 ml with H2O and
autoclave.
Solution 3: (This is also a concentrated stock)
NaCitrate 5.0g
MgSO4.7H2O
0.98g
(NH4)2SO4
8.98g
Dissolve and bring to 410 ml with H2O
Add 440 ml glycerol
Autoclave.
To make 1 liter of freezer medium, add
15 ml of solution 2 and 85 ml of solution 3 to the 900 ml of solution 1
and mix well. Add any required antibiotic before dispensing to microtiter
plates.
